From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) is designed for the in vitro quantitative determination of human Argininosuccinate Synthetase 1 (ASS1) in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich immunoassay technique, with an antibody specific for ASS1 pre-coated onto a microplate. Standards and samples are added to the wells and the ASS1 present in each sample is bound by the immobilised antibody. Following incubation and washing, the wells are incubated with a Biotinylated Anti-ASS1 Antibody that binds the captured ASS1. An HRP-Avidin conjugate is then added. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of ASS1 captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

The assay is quantitative, with a reported range of 1.56–100 ng/ml and a sensitivity of <0.63 ng/ml. It is run on a pre-coated microplate platform and takes 4 h 30 min. Kit components should be stored at +4 °C or –20 °C as instructed and not used past the expiration date.

Reported recovery averages are 91% for serum, 90% for EDTA plasma and 84% for heparin plasma, with the full set of recovery ranges provided for each. Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each of these sample types.
