Human Arachidonate Lipoxygenase 3 ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human e-LOX-3 (Arachidonate Lipoxygenase 3) in tissue homogenates, cell lysates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique using a 96-well microtiter plate pre-coated with an antibody specific for e-LOX-3. Standards and test samples are added to the wells, and e-LOX-3 present in each sample is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-e-LOX-3 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, generating a blue product that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader.

Quantitative performance characteristics include a sensitivity of <0.114 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Reported average recovery values across sample matrices were 91% for serum (range 80%–102%, n=5), 91% for EDTA plasma (range 81%–101%, n=5) and 84% for heparin plasma (range 80%–89%, n=5). Dilution linearity data for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions are also provided.

Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
