Human Arachidonate-5-Lipoxygenase ELISA Kit

Manufactured for Antibodies.com, this kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human 5 Lipoxygenase/5-LO in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Reactivity is reported as human, with detection performed on a colorimetric microplate reader.

The assay uses a pre-coated 96-well microplate (12 x 8 well strips) in which an immobilised antibody captures the target analyte from standards or test samples. Detection is carried out with a biotinylated anti-5 Lipoxygenase/5-LO antibody, followed by an HRP-avidin conjugate and TMB substrate; the resulting colour change is read at 450 nm and quantified against a standard curve. The reported standard range is 0.156-10 ng/ml with a sensitivity of 0.057 ng/ml, and total assay time is approximately 4 hours 30 minutes.

Spiked recovery values reported by the manufacturer (n=5) include serum averaging 91% (range 80-102%), EDTA plasma averaging 90% (range 81-100%) and heparin plasma averaging 84% (range 80-89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was observed for serum, EDTA plasma and heparin plasma samples. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
