Human Aquaporin 4 ELISA Kit

A sandwich ELISA from Antibodies.com designed for the quantitative in vitro determination of human Aquaporin 4 in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised antibody specific for Aquaporin 4. Standards and test samples are added to the wells, followed by washing and incubation with a Biotinylated Anti-Aquaporin 4 Antibody. An HRP-Avidin conjugate is then added, and TMB substrate solution is used to visualise the enzymatic reaction; the resulting yellow colour, whose intensity is proportional to the amount of Aquaporin 4 captured, is measured at 450 nm using a colorimetric microplate reader.

Reported sensitivity is <0.245 ng/ml, with an assay range of 0.625–40 ng/ml and a total assay time of 4h 30m. Recovery has been validated in serum (n=5, 80–102%, average 91%), EDTA plasma (n=5, 81–100%, average 90%), and heparin plasma (n=5, 80–89%, average 84%). Dilution linearity has been demonstrated in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, 1:8, and 1:16 dilutions.
