The microplate is pre-coated with an antibody specific for Aquaporin 2, also known by synonyms including AQP-2, AQP-CD, AQP2, Aquaporin-CD, ADH water channel, Collecting duct water channel protein, Water channel protein for renal collecting duct, and WCH-CD.

Standards and samples are added to the wells and bound by the immobilised antibody, followed by a biotinylated anti-Aquaporin 2 antibody and HRP-Avidin conjugate. The reaction is visualised with TMB substrate, with absorbance read at 450 nm. The assay has a sensitivity of <0.122 ng/ml and a working range of 0.312-20 ng/ml, with a total run time of 4 hours 30 minutes.

Sample recovery in serum averages 91% (range 80%-102%), in EDTA plasma averages 90% (range 81%-100%), and in heparin plasma averages 84% (range 80%-89%).
