Human Aquaporin 1, Colton Blood Group ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Aquaporin 1 in serum, plasma, tissue homogenates or other biological fluids. The assay targets Aquaporin 1, Colton Blood Group, and uses a pre-coated microplate onto which an antibody specific for Aquaporin 1 has been immobilised. Standards and test samples are added to the wells; following incubation and washing, a Biotinylated Anti-Aquaporin 1 Antibody is applied, followed by an HRP-Avidin conjugate. TMB substrate is then added to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Aquaporin 1 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

The kit has an assay range of 0.25–16 ng/ml and a sensitivity of <0.09 ng/ml. The assay is read on a colorimetric microplate reader and takes 4h 30m to complete. Reported recovery (n=5) is 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 (n=5) is 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
