Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Apolipoprotein D (Apo-D) in serum, plasma or other biological fluids, with readout on a colorimetric microplate reader.

The kit is supplied as a pre-coated microplate (12 x 8 well strips) with a total assay time of 4 hours 30 minutes. Reported sensitivity is <0.64 ng/ml and the assay range is 1.56–100 ng/ml. In the assay, an antibody specific for Apo-D is pre-coated onto the plate; standards and samples are captured by the immobilised antibody, then detected with a Biotinylated Anti-Apo-D Antibody followed by an HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of stop solution; absorbance is read at 450 nm and the concentration of Apo-D is calculated against the standard curve. Components include a Pre-Coated 96 Well Microplate (12 x 8 well strips), Lyophilized Standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers, with storage at +4°C or -20°C as instructed.

Spike-and-recovery linearity has been evaluated in serum, EDTA plasma and heparin plasma (n=5 each). Average recoveries are 91% (range 80–102%) in serum, 90% (range 81–100%) in EDTA plasma and 84% (range 80–89%) in heparin plasma. Dilution linearity has also been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for all three sample types.
