From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Apolipoprotein CII/ApoC-II in serum, plasma or other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) with an immobilised antibody specific for Apolipoprotein CII/ApoC-II. Standards and test samples are added, followed by a Biotinylated Anti-Apolipoprotein CII/ApoC-II Antibody and an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450nm in a microplate reader; the intensity of the yellow colour is proportional to the amount of Apolipoprotein CII/ApoC-II captured in each well.

The assay has a quantitative range of 3.12-200 ng/ml and a sensitivity of <1.53 ng/ml, with detection performed on a Colorimetric Microplate Reader. Total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

Spike-and-recovery performance has been validated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5) and heparin plasma (range 80%-89%, average 84%, n=5). Dilution linearity has also been tested at 1:2, 1:4, 1:8 and 1:16 dilutions across serum (87-91% to 92-97% across dilutions), EDTA plasma (90-105% to 79-108%) and heparin plasma (84-95% to 89-91%).
