Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Apolipoprotein B in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colorimetric detection read at 450 nm on a microplate reader. Standards and samples are added to the wells, where Apolipoprotein B is captured by the immobilized antibody, followed by incubation with a biotinylated anti-Apolipoprotein B antibody and an HRP-Avidin conjugate. TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of Apolipoprotein B captured in each well.
The quantitative sandwich assay has a reported product range of 12.5–800 pg/ml and a sensitivity of less than 5.9 pg/ml, with a total assay time of approximately 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed in the protocol manual, and the kit must not be used past its expiration date.
