From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Apelin 17 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The 96-well microtiter plate is pre-coated with Apelin 17 antigen. During incubation, Apelin 17 present in the samples or standards competes with the fixed amount of immobilized Apelin 17 for binding sites on the Biotinylated Anti-Apelin 17 Antibody. Following incubation, unbound antibody is removed by washing, and an HRP-Avidin conjugate is added. A TMB substrate solution is then used to visualize the enzymatic reaction, producing a blue product that changes to yellow after addition of acidic stop solution. The density of yellow is inversely proportional to the amount of Apelin 17 present, and the concentration can be calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The standard product range is 0.156–10 ng/ml with a sensitivity of 0.072 ng/ml. The assay can be completed in 2h 30m using a colorimetric microplate reader, and the platform is a pre-coated microplate comprising 12 x 8 well strips. Reported recovery averages are: serum 91% (range 83–100%), EDTA plasma 96% (range 90–103%), and heparin plasma 95% (range 89–102%), with linearity demonstrated across dilutions of 1:2, 1:4, 1:8 and 1:16 in the same matrices.

Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
