This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Anterior Gradient Protein 2 (AGR2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for AGR2 has been pre-coated onto a 96-well microplate. Standards and test samples are added to the wells; AGR2 present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-AGR2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualize the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. The density of yellow is proportional to the amount of AGR2 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

The assay has a product range of 0.156-10 ng/ml and a sensitivity of 0.061 ng/ml. It is read on a Colorimetric Microplate Reader, with a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed. Spike-and-recovery testing (n=5) yielded averages of 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). Dilution linearity across 1:2 to 1:16 dilutions was demonstrated for serum, EDTA plasma, and heparin plasma samples.
