Human Anoctamin 6 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human ANO6 (Anoctamin 6) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate immobilised with an antibody specific for ANO6. Standards and samples are added to the wells, followed by a biotinylated anti-ANO6 detection antibody and an HRP-avidin conjugate. TMB substrate is then added; the resulting yellow product, measured at 450 nm, is proportional to ANO6 concentration and is quantified against a standard curve.

The assay has a reported range of 0.312–20 ng/ml and a sensitivity of <0.127 ng/ml, with a total run time of 4 hours 30 minutes. Detection is read on a colorimetric microplate reader. The kit has been validated with serum, EDTA plasma, and heparin plasma samples. Reported average recovery values across sample sets of 5 are 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%), and 84% for heparin plasma (range 80%–89%). Dilutional linearity across 1:2 to 1:16 sample dilutions was also characterised for each matrix.

Supplied components include a pre-coated microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, with storage temperatures specified for each reagent. The kit should not be used past its expiration date.
