Human Angiopoietin 1 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Angiopoietin 1 in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for Angiopoietin 1 pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells, and Angiopoietin 1 is captured by the immobilised antibody before incubation with a Biotinylated Anti-Angiopoietin 1 Antibody and HRP-Avidin conjugate. TMB substrate is then added, producing a blue-coloured product that turns yellow on addition of acidic stop solution; absorbance is read at 450 nm in a colourmetric microplate reader.

The assay has a reported sensitivity of <0.059 ng/ml and a quantitative range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Sample recovery data are reported for serum (average 91%, range 80–102%, n=5), EDTA plasma (average 91%, range 81–101%, n=5) and heparin plasma (average 84%, range 80–89%, n=5), with supporting linear dilution performance reported across 1:2 to 1:16 for each matrix.

Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A and Detection Solution B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or -20°C as instructed and the kit is for use with a colourmetric microplate reader.
