Human Amyloid Beta Peptide 1-40 ELISA Kit

Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is intended for the in vitro quantitative determination of human Amyloid beta Peptide 1-40 in serum, plasma or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with Amyloid beta Peptide 1-40 antigen. During incubation, Amyloid beta Peptide 1-40 in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-Amyloid beta Peptide 1-40 Antibody. An HRP-Avidin conjugate is then added, and detection is performed with a TMB substrate solution that produces a blue product turning yellow after the addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader, with the density of yellow colour inversely proportional to the amount of analyte present.

The assay has a working range of 15.6–1000 pg/ml and a sensitivity of 5.58 pg/ml. Total assay time is approximately 2 hours 30 minutes. The pre-coated microplate is configured as 12 × 8 well strips, and reagent components are stored at +4°C or –20°C as instructed; the kit should not be used past its expiration date.

Validation data for sample recovery (n = 5) report averages of 91% in serum (range 83–100%), 95% in EDTA plasma (range 91–99%) and 94% in heparin plasma (range 90–99%).
