Human Amylase Alpha 1, Salivary ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, intended for the in vitro quantitative determination of human Salivary alpha Amylase in serum, plasma, saliva or other biological fluids, with reactivity against human samples.

The assay employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips. An antibody specific for Salivary alpha Amylase is immobilised on the wells; standards and test samples are added and the analyte is captured by the immobilised antibody. After washing, a Biotinylated Anti-Salivary alpha Amylase Antibody binds the captured analyte, followed by addition of HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of Salivary alpha Amylase is calculated from the standard curve. Detection is colorimetric.

Working range is 3.12-200 ng/ml with a sensitivity of <1.32 ng/ml, and total assay time is 4h 30m. Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers, stored at +4°C or -20°C as instructed. Average recovery values are 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%) and 84% for heparin plasma (range 80%-89%).
