Description

This Antibodies.com ELISA kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human SCNN1G in tissue homogenates or other biological fluids. The assay is reactive with human samples and targets the Amiloride Sensitive Sodium Channel Subunit Gamma protein.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for SCNN1G is pre-coated onto a microtiter plate. Standards and test samples are added to the wells and SCNN1G present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-SCNN1G Antibody is added, followed by HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The concentration of SCNN1G is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

Reported sensitivity is <0.113 ng/ml with an assay range of 0.312-20 ng/ml, and a total assay time of 4h 30m. Average recovery values in spiked samples are 91% for serum (n=5, range 83-100%), 94% for EDTA plasma (n=5, range 90-98%), and 95% for heparin plasma (n=5, range 89-102%).
