A competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com designed for the in vitro quantitative determination of human alpha-Melanocyte Stimulating Hormone (alpha MSH). The assay is reactive with human samples.

The kit employs a competitive enzyme immunoassay technique in which alpha MSH present in standards or samples competes with alpha MSH pre-coated on the microplate for binding sites on a Biotinylated Anti-alpha MSH Antibody. After washing, an HRP-Avidin conjugate is added, and TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, with yellow intensity inversely proportional to the alpha MSH concentration. Sensitivity is <6.43 pg/ml and the product range is 15.6-1000 pg/ml. Total assay time is 2h 30m.

Validated sample types include serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Reported recovery values (n=5) average 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%), and 84% for heparin plasma (range 80%-89%). Linearity has been demonstrated across serial dilutions (1:2, 1:4, 1:8, 1:16) for serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the product must not be used past its expiration date.
