Human Alkaline Phosphatase, Placental Like Protein 2 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human ALPPL2 (Alkaline Phosphatase, Placental Like Protein 2) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for ALPPL2 is immobilised on the plate; standards and samples are added, followed by a biotinylated anti-ALPPL2 detection antibody and an HRP-avidin conjugate. The reaction is visualised using TMB substrate, producing a blue product that turns yellow upon addition of stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

The assay has a measuring range of 0.312–20 ng/ml and a sensitivity of <0.113 ng/ml, with a total assay time of 4 hours 30 minutes. Spike-and-recovery validation reports average recoveries of 91% in serum (range 80–102%, n=5), 91% in EDTA plasma (range 81–101%, n=5), and 84% in heparin plasma (range 80–89%, n=5). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 across all three sample matrices.

Components supplied include the pre-coated microplate, lyophilised standard (2 vials), detection solutions A and B (120 µl each), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and five adhesive plate sealers. Kit components are stored at +4 °C or –20 °C as instructed, and the kit should not be used past its expiration date.
