Human Aldose Reductase Like Protein 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human AKR1B10 (Aldose Reductase Like Protein 1) in serum, plasma, tissue homogenates or other biological fluids.

The assay employs a pre-coated microplate to which an antibody specific for AKR1B10 is immobilised. Standards and test samples are added and AKR1B10 present is bound by the immobilised antibody. After incubation and washing, a Biotinylated Anti-AKR1B10 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and AKR1B10 concentration is calculated from a standard curve.

Key performance characteristics include a sensitivity of <0.064 ng/ml and an assay range of 0.156–10 ng/ml. Total assay time is 4 h 30 min. Detection is colourimetric on a pre-coated 96-well microplate (12 x 8 well strips). Kit components are stored at +4°C or -20°C as instructed. Sample data (n=5) showed averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Across dilution factors of 1:2, 1:4, 1:8, and 1:16 (n=5), values ranged from 74–107% for serum, 79–108% for EDTA plasma, and 82–105% for heparin plasma.
