Description
A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Aldolase (Aldolase A, Fructose Bisphosphate) in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique in which an antibody specific for Aldolase is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, the wells are washed, and detection is carried out using a Biotinylated Anti-Aldolase Antibody followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. The density of yellow is proportional to the amount of Aldolase captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450nm on a colorimetric microplate reader and referring to the standard curve.
The reported assay range is 62.5-4000 pg/ml, with a sensitivity of <29.3 pg/ml. The full assay can be completed in 4h 30m. Manufacturer-reported spike-and-recovery values across n=5 samples include serum 80%-102% (average 91%), EDTA plasma 81%-100% (average 90%) and heparin plasma 80%-89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma, each across n=5 samples.
Reactivity is Human. Kit components include the pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Storage instructions call for component-specific temperatures of +4°C or -20°C as indicated, and the kit is not to be used past its expiration date.
