Human Aggrecan ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Aggrecan in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate (12 x 8 well strips). An antibody specific for Aggrecan is immobilised on the plate; standards and test samples are added, followed by a Biotinylated Anti-Aggrecan Antibody and an HRP-Avidin conjugate. After incubation and washing, TMB substrate solution produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a microplate reader.

The colourimetric, quantitative assay has a reported range of 0.468-30 ng/ml and a sensitivity of <0.16 ng/ml, with a total assay time of 4h 30m. In spike-and-recovery testing, recoveries were 80%-102% (average 91%) in serum (n=5), 81%-100% (average 90%) in EDTA plasma (n=5) and 80%-89% (average 84%) in heparin plasma (n=5). Dilution linearity data report ranges such as 87-107% for serum at 1:4 and 84-101% for EDTA plasma at 1:4, across dilutions tested from 1:2 to 1:16.

Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date. The kit includes a pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, along with an easy-to-follow protocol.
