The Antibodies.com Human Adrenomedullin Receptor ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human GPR182 in tissue homogenates, cell lysates, or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for GPR182 is pre-coated onto a 96-well microplate, standards and samples are added, and GPR182 present in each sample is bound by the immobilised antibody. After incubation and washing, a Biotinylated Anti-GPR182 Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution then visualises the HRP enzymatic reaction as a blue-coloured product that turns yellow upon addition of acidic stop solution. The concentration of GPR182 is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Key specifications include a sensitivity of <0.049 ng/ml, an assay range of 0.156-10 ng/ml, and a total assay time of 4h 30m. The assay is run on a pre-coated microplate platform (12 x 8 well strips) with colourimetric detection. Validation data report recovery in serum (n=5) at 80-102% with an average of 91%, in EDTA plasma (n=5) at 81-100% with an average of 90%, and in heparin plasma (n=5) at 80-89% with an average of 84%, together with dilution linearity data across 1:2, 1:4, 1:8 and 1:16 sample dilutions. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
