A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human PACAP-38 (Adenylate Cyclase Activating Polypeptide 1, Pituitary) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, a biotinylated anti-PACAP-38 detection antibody, HRP-avidin and TMB substrate, with absorbance read at 450 nm. Total assay time is 4 h 30 m and components are stored at +4 °C or −20 °C as indicated.

Performance characteristics include a standard curve range of 7.81–500 pg/ml and a stated sensitivity of <2.3 pg/ml. Spike-and-recovery in serum (n=5) averaged 91% (range 80–102%), in EDTA plasma 91% (81–101%) and in heparin plasma 84% (80–89%). Dilution linearity across 1:2 to 1:16 was 87–107% for serum, 84–108% for EDTA plasma and 82–105% for heparin plasma. The kit is supplied with lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
