A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Adenylate Cyclase 1/AC1 in tissue homogenates, cell lysates or other biological fluids. The kit uses a pre-coated 96-well microplate platform (12 x 8 well strips) with a colourmetric readout measured at 450 nm in a microplate reader. The assay covers a product range of 0.781–50 ng/ml with a sensitivity of <0.28 ng/ml, and the full protocol takes approximately 4 h 30 m.

An antibody specific for Adenylate Cyclase 1/AC1 is immobilised on the plate; standards and test samples are added and the analyte is bound by the capture antibody. After washing, a Biotinylated Anti-Adenylate Cyclase 1/AC1 antibody binds the captured target, followed by an HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow upon addition of the acidic stop solution; the absorbance density is proportional to Adenylate Cyclase 1/AC1 concentration and is quantified against a standard curve.

Recovery was evaluated across sample matrices. Average recovery in serum was 91% (range 80%–102%, n = 5), in EDTA plasma 90% (range 81%–100%, n = 5), and in heparin plasma 84% (range 80%–89%, n = 5). Dilution linearity was also assessed at 1:2, 1:4, 1:8 and 1:16 across the same matrices. Kit components should be stored at +4 °C or −20 °C as instructed and not used past the expiration date.
