From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human alpha Actinin 4 in tissue homogenates, cell lysates and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for alpha Actinin 4; standards and samples are added so that the target analyte is captured by the immobilised antibody. After washing, a biotinylated anti-alpha Actinin 4 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of alpha Actinin 4 captured in each well, and the concentration is calculated from the O.D. absorbance at 450 nm against a standard curve.
Reported sensitivity is <0.24 ng/ml with an assay range of 0.781–50 ng/ml. Total assay time is 4 hours 30 minutes, and detection is colorimetric on a pre-coated microplate comprising 12 x 8 well strips. The kit is reactive for human samples and is intended for quantitative measurement in cell lysates and tissue homogenates. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
