Human Actinin Alpha 3 ELISA Kit

A sandwich ELISA kit from Antibodies.com designed for the in vitro quantitative determination of human Actinin Alpha 3 (ACTN3) in tissue homogenates, cell lysates, and other biological fluids. The assay uses a colorimetric detection method, with absorbance read on a microplate reader at 450 nm.

The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for ACTN3 pre-coated onto a microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-ACTN3 Antibody and an HRP-Avidin conjugate. A TMB substrate solution is then added; catalysis produces a blue-coloured product that changes to yellow after the addition of acidic stop solution. The density of yellow is proportional to the amount of ACTN3 captured, and concentration is calculated by reference to a standard curve. Sensitivity is quoted at 0.115 ng/ml with an assay range of 0.312–20 ng/ml, and the complete protocol takes 4 hours 30 minutes.

Recovery has been validated for serum (range 80%–102%, average 91%), EDTA plasma (range 81%–100%, average 90%) and heparin plasma (range 80%–89%, average 84%). Kit components are stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
