Description

Manufactured for Antibodies.com, this assay is a sandwich ELISA (sELISA) designed for the in vitro quantitative determination of human Sarcomeric alpha Actinin (Actinin Alpha 2) in tissue homogenates or other biological fluids. The kit is intended for human samples and is classified under Assays and Kits.

The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Sarcomeric alpha Actinin is immobilised on the plate; standards and test samples are added, and any target present is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-Sarcomeric alpha Actinin Antibody binds the captured target, and an HRP-Avidin conjugate is then added. TMB substrate solution is used to visualise the HRP enzymatic reaction as a blue product that turns yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of target captured and is measured by O.D. absorbance at 450 nm using a colorimetric microplate reader. Total assay time is 4 hours 30 minutes.

Performance specifications include a sensitivity of <0.112 ng/ml and an assay range of 0.312-20 ng/ml. Recovery data (n=5) are reported for serum (range 80%-102%, average 91%), EDTA plasma (range 81%-100%, average 90%), and heparin plasma (range 80%-89%, average 84%), with linearity data provided across dilutions from 1:2 to 1:16 for all three sample types. Kit components should be stored at +4°C or -20°C as instructed, and components should not be used past the expiration date.
