The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Acid Sphingomyelinase pre-coated onto a 96-well microplate. Standards and samples are added to the wells, and bound Acid Sphingomyelinase is detected using a Biotinylated Anti-Acid Sphingomyelinase Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a colour change that is read at 450 nm on a colorimetric microplate reader.

The reported product range is 0.312-20 ng/ml with a sensitivity of 0.117 ng/ml, and the assay can be completed in 4h 30m. Recovery in serum averages 91% (range 80%-102%), in EDTA plasma 90% (range 81%-100%), and in heparin plasma 84% (range 80%-89%).
