Human Acetylcholinesterase Associated Protein ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of human CutA in tissue homogenates and other biological fluids. The pre-coated microplate is supplied with antibody specific for CutA immobilised on the wells. Standards and test samples are added to the wells, and any CutA present is bound by the immobilised antibody. After washing, a biotinylated anti-CutA detection antibody is added, followed by an HRP-avidin conjugate. The reaction is visualised using a TMB substrate, producing a blue colour that turns yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of CutA captured in each well, and the concentration is calculated by reading the absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The assay has a working range of 0.156–10 ng/ml and a sensitivity of <0.051 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery has been validated in serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5) and heparin plasma (range 80–89%, average 84%, n=5). Linearity has been demonstrated across dilutions of 1:2, 1:4, 1:8 and 1:16 for serum (87–91%, 87–107%, 74–101% and 92–97% respectively), EDTA plasma (90–105%, 84–101%, 90–101% and 79–108%) and heparin plasma (84–95%, 92–105%, 82–105% and 89–91%). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
