The kit targets Acetyl Coenzyme A Acetyltransferase 2 and is reactive with human samples, making it suitable for cell lysate and tissue homogenate analysis.

The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate. An antibody specific for ACAT2/Acetyl-CoA Acetyltransferase immobilises the analyte from standards and test samples; after washing, a Biotinylated Anti-ACAT2/Acetyl-CoA Acetyltransferase Antibody binds the captured target. Following incubation with HRP-Avidin conjugate and further washing, TMB substrate is added to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the analyte concentration, which is calculated by reading the absorbance at 450 nm in a colorimetric microplate reader against a standard curve. The complete assay time is 4 h 30 min.

The kit delivers a quantitative measurement range of 78.1–5000 pg/ml with a sensitivity of 31.5 pg/ml. Validation studies report the following average recovery values (n=5) across undiluted samples: serum 91% (range 80–102%), EDTA plasma 90% (range 81–100%), and heparin plasma 84% (range 80–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions, yielding overall recovery ranges of 74–107% for serum, 79–108% for EDTA plasma, and 82–105% for heparin plasma.
