From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is designed for the in vitro quantitative determination of human ADAMTS5 in tissue homogenates, cell lysates, or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for ADAMTS5 is pre-coated onto a 96-well microplate (12 x 8 well strips). Standards and test samples are added to the wells, where ADAMTS5 is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-ADAMTS5 Antibody binds the captured ADAMTS5 in each well. An HRP-Avidin conjugate is then added, and after further incubation and washing, TMB substrate solution visualises the HRP enzymatic reaction to yield a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of ADAMTS5 captured. The concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Specifications include a sensitivity of <0.132 ng/ml, an assay range of 0.312–20 ng/ml, an assay time of 4h 30m, and colorimetric detection on a pre-coated microplate. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date. The kit is suitable for cell lysates and tissue homogenates. Reported average recovery in serum (n=5) is 91% (range 80%–102%), in EDTA plasma (n=5) is 91% (range 81%–101%), and in heparin plasma (n=5) is 84% (range 80%–89%).
