The assay uses a 96-well microplate pre-coated with an antibody specific for ADAM17. Standards and test samples are added to the wells and bound by the immobilised antibody, followed by a Biotinylated Anti-ADAM17 Antibody and an HRP-Avidin conjugate. A TMB substrate generates a colour change that is read at 450 nm on a colorimetric microplate reader, with absorbance proportional to ADAM17 concentration and quantified against a standard curve.

The kit has an assay range of 0.156–10 ng/ml and a sensitivity of <0.056 ng/ml. Total assay time is approximately 4 hours 30 minutes.
