Human A Disintegrin And Metalloprotease 15 ELISA Kit

This sandwich ELISA kit, available through Antibodies.com, is designed for the in vitro quantitative determination of human ADAM15 (A Disintegrin And Metalloprotease 15) in serum, plasma, tissue homogenates, and other biological fluids. The assay employs an antibody specific for ADAM15 pre-coated onto a 96-well microplate; standards and test samples are added and ADAM15 present in each sample is bound to the wells by the immobilised antibody. After washing, a biotinylated anti-ADAM15 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a yellow product after addition of the acidic stop solution; absorbance is read at 450 nm in a microplate reader and the concentration of ADAM15 is calculated against the standard curve.

Performance characteristics reported by the manufacturer include a sensitivity of <0.054 ng/ml and an assay range of 0.156–10 ng/ml. Recovery of spiked ADAM15 averaged 91% (range 83–100%) in serum, 94% (range 90–98%) in EDTA plasma, and 95% (range 89–102%) in heparin plasma (n=5 per sample type). Dilution linearity across 1:2 to 1:16 sample dilutions was reported as 83–110% for serum, 73–109% for EDTA plasma, and 77–110% for heparin plasma. The total assay time is 4 hours 30 minutes, and kit components are stored at +4°C or –20°C as instructed; the kit should not be used past its expiration date.
