This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of human ADAM12 (A Disintegrin And Metalloprotease 12) in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated microplate platform and follows the sandwich enzyme immunoassay principle: an antibody specific for ADAM12 is immobilised on the plate, standards and samples are added, and captured analyte is detected with a Biotinylated Anti-ADAM12 Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate and stopped with acidic stop solution, producing a yellow product that is read at 450 nm on a colorimetric microplate reader; absorbance is proportional to ADAM12 concentration and is quantified against a standard curve.

The reported sensitivity is <28 pg/ml and the standard curve covers an assay range of 78.1–5000 pg/ml. Total assay time is 4 hours 30 minutes. Validation data supplied by the manufacturer include mean recoveries of 91% for serum (n=5, range 83–100%), 94% for EDTA plasma (n=5, range 90–98%) and 95% for heparin plasma (n=5, range 89–102%), together with dilution linearity results across 1:2, 1:4, 1:8 and 1:16 dilutions for each of the three sample types. Kit components include the pre-coated microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer (30X concentrate), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers; components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
