This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human ADAM10 in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for ADAM10 pre-coated onto a microplate. Standards and samples are added to the wells, where ADAM10 is bound by the immobilized antibody. After washing, a Biotinylated Anti-ADAM10 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of stop solution, and absorbance is read at 450nm in a microplate reader.

Key specifications include a sensitivity of less than 24.6 pg/ml and a working range of 62.5–4000 pg/ml. Detection is colorimetric on a pre-coated 12 x 8 well strip microplate, with a total assay time of 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed.

Spike-and-recovery testing demonstrated reliable performance across sample types: serum (n=5) showed 83%–100% recovery with a 91% average, EDTA plasma (n=5) showed 90%–98% recovery with a 94% average, and heparin plasma (n=5) showed 89%–102% recovery with a 95% average.
