Human 3-Hydroxybutyrate Dehydrogenase 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human 3-Hydroxybutyrate Dehydrogenase 1 (BDH1) in tissue homogenates, cell lysates and other biological fluids. The kit is reactive with human samples and employs the sandwich immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips, read on a colorimetric microplate reader.

Performance characteristics include a sensitivity of <0.055 ng/ml and a quantitative assay range of 0.156–10 ng/ml. The complete assay runs in approximately 4 hours 30 minutes, with colourmetric detection by measuring absorbance at 450 nm. Kit components are stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.

In the assay workflow, standards and test samples are added to wells where BDH1 is captured by the immobilised antibody. Wells are washed and incubated with a Biotinylated Anti-BDH1 Antibody, followed by HRP-Avidin conjugate. TMB substrate solution produces a blue-coloured product that turns yellow upon addition of acidic stop solution; the intensity of yellow colour is directly proportional to the amount of BDH1 captured in each well, allowing concentration to be calculated by reference to the standard curve.
