A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of guinea pig Visfatin in serum, plasma, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Visfatin that is pre-coated onto a microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-Visfatin Antibody and an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow after addition of the acidic stop solution. The density of yellow is proportional to the amount of Visfatin captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The kit reports a sensitivity of <0.52 ng/ml and an assay range of 1.56–100 ng/ml, with a total assay time of 4 h 30 min. Kit components are stored at +4 °C or –20 °C as instructed and the kit should not be used past its expiration date. Average recovery values (n=5) are reported as: serum 80–102 % (average 91 %), EDTA plasma 81–99 % (average 90 %), and heparin plasma 80–89 % (average 84 %). Dilution linearity ranges at 1:2, 1:4, 1:8 and 1:16 dilutions are reported for serum (n=5) as 87–91 %, 87–107 %, 74–101 % and 92–97 %; for EDTA plasma (n=5) as 90–105 %, 84–101 %, 90–101 % and 79–108 %; and for heparin plasma (n=5) as 84–95 %, 92–105 %, 82–105 % and 89–91 %.
