Guinea Pig Matrix Metalloproteinase 8 ELISA Kit

The kit employs a pre-coated 96-well microtiter plate to which an antibody specific for MMP8 has been immobilised. Standards and test samples are added to the wells; following incubation and washing, a biotinylated anti-MMP8 antibody binds the captured MMP8, HRP-Avidin conjugate is added, and TMB substrate solution is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution.

Absorbance is read at 450 nm on a colorimetric microplate reader, with MMP8 concentration calculated by reference to a standard curve. The quantitative sandwich assay reports a sensitivity of 0.072 ng/ml, a product range of 0.156-10 ng/ml, and a total assay time of 4h 30m. Reported recovery values are 80-102% (average 91%) in serum (n=5), 81-100% (average 90%) in EDTA plasma (n=5), and 80-89% (average 84%) in heparin plasma (n=5). Dilution linearity data is provided across 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma samples.
