It is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids and is read on a colorimetric microplate reader measuring absorbance at 450 nm. The 96-well microplate is pre-coated with anti-IL-10 capture antibody; bound IL-10 is detected with a biotinylated anti-IL-10 antibody, an HRP-avidin conjugate and TMB substrate, producing a yellow product on addition of stop solution whose absorbance is proportional to IL-10 concentration and quantified against the standard curve.

Quoted performance figures include a product range of 15.6-1000 pg/ml, a sensitivity of <5.2 pg/ml and a total assay time of 4h 30m. Spiked recovery testing (n=5) gave average recoveries of 91% in serum, 96% in EDTA plasma and 95% in heparin plasma. Dilution linearity across 1:2 to 1:16 (n=5) was reported between 79% and 110% in serum, between 73% and 109% in EDTA plasma, and between 77% and 110% in heparin plasma.
