Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of guinea pig Interferon Gamma in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for Interferon Gamma is pre-coated onto a 96-well microtiter plate, and standards and test samples are added so that the target in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-Interferon Gamma Antibody binds the captured target, followed by an HRP-Avidin conjugate. TMB substrate solution then visualises the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of an acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of Interferon Gamma is calculated by reference to a standard curve.

The reported product range is 15.6-1000 pg/ml with a sensitivity of 6.3 pg/ml, and the assay is completed in approximately 4 hours 30 minutes on a pre-coated microplate (12 x 8 well strips). Kit components include a Pre-Coated 96 Well Microplate, Lyophilised Standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and Plate Sealers, stored at +4°C or -20°C as instructed. Reported average recovery ranges are 80% - 102% for serum, 81% - 100% for EDTA plasma and 80% - 89% for heparin plasma, with corresponding mean recoveries of 91%, 90% and 84%. Reported linearities across 1:2, 1:4, 1:8 and 1:16 dilutions are 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
