Description

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of goat Bone Morphogenetic Protein 4 (BMP4) in serum, plasma, tissue homogenates and other biological fluids. The assay employs a pre-coated 96-well microplate configured as 12 x 8 well strips, with an antibody specific for BMP4 immobilised on the plate. Standards and test samples are added to the wells, followed by a biotinylated anti-BMP4 antibody and an HRP-avidin conjugate. TMB substrate is then added and the enzymatic reaction is visualised by catalysis, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The density of the yellow colour is proportional to the amount of BMP4 captured in each well, and the concentration is calculated from the O.D. absorbance read at 450 nm on a colorimetric microplate reader against a standard curve.

The quantitative assay range is 0.312–20 ng/ml with a reported sensitivity of <0.148 ng/ml, and the complete protocol takes approximately 4h 30m. Recovery validation across serum, EDTA plasma and heparin plasma (n=5 each) returned average recoveries of 91%, 90% and 84% respectively, with corresponding ranges of 80%–102%, 81%–100% and 80%–89%. Linearity was confirmed across serial dilutions from 1:2 to 1:16 for serum (87–107%), EDTA plasma (79–108%) and heparin plasma (82–105%). Kit components are stored at +4°C or −20°C as instructed and the kit should not be used past its expiration date.
