Description

A competitive Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of goat adrenocorticotropic hormone (ACTH) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The kit employs the competitive enzyme immunoassay technique on a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips, where ACTH in samples or standards competes with immobilized ACTH for binding sites on a Biotinylated Anti-ACTH Antibody. After incubation and washing, an HRP-Avidin conjugate is added and TMB substrate is used to visualise the reaction. Absorbance is read at 450 nm in a colorimetric microplate reader, with the density of yellow inversely proportional to the amount of ACTH present. Total assay time is 2h 30m.

Quantitative performance: the product range is 15.6-1000 pg/ml with a sensitivity of 5.15 pg/ml. Reported sample recovery values across five replicates were 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 (n=5) gave 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
