Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of Progesterone in serum, plasma, or other biological fluids. Reactivity is described as universal, and the assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips.

The assay uses a competitive enzyme immunoassay technique. Progesterone in samples or standards competes with a fixed amount of immobilized Progesterone antigen for binding sites on a biotinylated anti-progesterone antibody. After washing, an HRP-avidin conjugate is added, followed by TMB substrate and an acidic stop solution that produces a yellow colour. Optical density is read at 450 nm in a colorimetric microplate reader, and the yellow intensity is inversely proportional to the Progesterone concentration, which is calculated from a standard curve.

The manufacturer reports a sensitivity of <0.061 ng/ml, an assay range of 0.156-10 ng/ml, and a total assay time of 2h 30m. Reported values across n=5 samples are 80%-102% (average 91%) in serum, 81%-99% (average 90%) in EDTA plasma, and 80%-89% (average 84%) in heparin plasma. Values are also provided for dilutions of 1:2, 1:4, 1:8, and 1:16: serum (n=5) 87-91%, 87-107%, 74-101%, and 92-97%; EDTA plasma (n=5) 90-105%, 84-101%, 90-101%, and 79-108%; heparin plasma (n=5) 84-95%, 92-105%, 82-105%, and 89-91%. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
