This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of Ethinylestradiol in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. It provides universal reactivity and is intended for quantitative measurement across a broad range of sample matrices including serum, EDTA plasma, and heparin plasma.

The assay uses the competitive enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a Biotinylated Anti-Ethinylestradiol Antibody and an HRP-Avidin conjugate. Bound analyte is visualised with TMB substrate, producing a yellow colour after addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader. The standard curve covers a product range of 31.2–2000 pg/ml, with a sensitivity of 9.48 pg/ml, and the total assay time is approximately 2 h 30 m. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

Recovery and dilution linearity were verified for serum, EDTA plasma, and heparin plasma. In serum (n=5), recovery ranged from 80%–102% with an average of 91%; in EDTA plasma (n=5), 81%–99% with an average of 90%; and in heparin plasma (n=5), 80%–89% with an average of 84%. Dilution linearity at 1:2, 1:4, 1:8, and 1:16 dilutions was 87–91%, 87–107%, 74–101%, and 92–97% respectively for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma.
