General Estrone Sulfate ELISA Kit

From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of Estrone Sulfate in serum, plasma or other biological fluids. The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with the target antigen pre-bound to the plate wells.

During the assay, Estrone Sulfate in samples or standards competes with the immobilized antigen for binding sites on a Biotinylated Anti-Estrone Sulfate Antibody. After washing, an HRP-Avidin conjugate is added, followed by TMB substrate. The reaction produces a blue colour that turns yellow upon addition of stop solution, with absorbance read at 450 nm. The yellow intensity is inversely proportional to the Estrone Sulfate concentration, allowing quantification against a standard curve.

The assay has a working range of 1.56-100 ng/ml with a sensitivity of <0.51 ng/ml, and a total assay time of 2h 30m. Recovery has been validated in serum (83%-100%, average 91%, n=5), EDTA plasma (90%-103%, average 96%, n=5), and heparin plasma (89%-102%, average 95%, n=5). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 ratios across the same sample types. Kit components are stored at +4°C or -20°C as instructed.
