General Estrone ELISA Kit

Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) kit is designed for the in vitro quantitative determination of Estrone in serum, plasma, or other biological fluids. The kit employs a pre-coated 96-well microplate arranged as 12 x 8 well strips and is intended for use with a colorimetric microplate reader, with absorbance measured at 450 nm. The quantitative assay range is 125-8000 pg/ml, with a sensitivity of <62.4 pg/ml. Total assay time is 2 hours 30 minutes, and kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

The assay uses the competitive enzyme immunoassay technique, in which Estrone present in samples or standards competes with immobilized Estrone antigen for binding sites on a biotinylated anti-Estrone antibody. After washing, an HRP-avidin conjugate is added to the wells, followed by TMB substrate solution, which is catalysed to produce a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow colour is inversely proportional to the amount of Estrone present in each sample or standard, and concentration is calculated by referring to the standard curve.

Reported spike-and-recovery performance with n=5 per matrix was: serum 80%-102% (average 91%), EDTA plasma 81%-99% (average 90%), and heparin plasma 80%-89% (average 84%). Dilution linearity data are also provided for serum, EDTA plasma, and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16.
