From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of Dihydrotestosterone in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay is run on a pre-coated 96-well microplate (12 x 8 well strips) and read colorimetrically at 450 nm in a microplate reader.
The standard curve covers 31.2–2000 pg/ml with a sensitivity of 16.15 pg/ml, and the total assay time is 2 hours 30 minutes. In the competitive format, Dihydrotestosterone in samples or standards competes with immobilised antigen for binding to a biotinylated anti-Dihydrotestosterone antibody; HRP-Avidin and TMB substrate are used for detection, producing a yellow colour whose intensity is inversely proportional to the amount of analyte present.
Validation data reported for serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5) include average recoveries of 91% (range 80–102%), 90% (range 81–100%) and 84% (range 80–89%), respectively. Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 across the same sample types. Kit components are stored at +4°C or −20°C as instructed and should not be used past the expiration date.
