General Cyclic Guanosine Monophosphate ELISA Kit

Manufactured by Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of cyclic guanosine monophosphate (cGMP) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids.

The assay uses a pre-coated microplate to which cGMP antigen is immobilised. During incubation, cGMP present in samples or standards competes with the fixed antigen for binding sites on a biotinylated anti-cGMP antibody. After washing to remove unbound antibody, an HRP-avidin conjugate is added and incubated. A TMB substrate solution then visualises the HRP enzymatic reaction by catalysis, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The density of yellow is inversely proportional to the amount of cGMP in each sample or standard; concentration is calculated by reading the O.D. absorbance at 450 nm on a microplate reader and referring to the standard curve.

The kit has a stated sensitivity of <0.061 ng/ml within a measurable range of 0.156–10 ng/ml, with a total assay time of approximately 2 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed, and the assay is performed on a colorimetric microplate reader.
