A competitive Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of cyclic adenosine monophosphate (cAMP) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The kit employs a competitive enzyme immunoassay technique using a pre-coated microplate, where cAMP present in samples or standards competes with the immobilised cAMP antigen for binding sites on a Biotinylated Anti-cAMP Antibody. Following incubation and washing, an HRP-Avidin conjugate is added, and a TMB substrate solution is used to visualise the enzymatic reaction, producing a colour change that is inversely proportional to the amount of cAMP present. Absorbance is read at 450 nm using a colorimetric microplate reader, with the cAMP concentration calculated by reference to a standard curve.

The assay has a working range of 0.312-20 ng/ml and a sensitivity of <0.118 ng/ml, with a total run time of 2h 30m. Recovery testing yielded ranges of 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma. Linearity was demonstrated at dilutions of 1:2, 1:4, 1:8, and 1:16 across the same three sample types. Kit components include a lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Storage requirements vary by component between +4°C and -20°C, and the kit should not be used past its expiration date.
