A competitive enzyme-linked immunosorbent assay (cELISA) from Antibodies.com enables the in vitro quantitative determination of Cortisone in serum, plasma, urine or other biological fluids. The assay uses a pre-coated microplate in which Cortisone present in samples or standards competes with immobilised antigen for binding sites on a biotinylated anti-Cortisone antibody. Detection is achieved using an HRP-avidin conjugate and TMB substrate, with absorbance read at 450 nm on a colorimetric microplate reader.
The manufacturer states an assay range of 3.12-200 ng/ml and a sensitivity of 0.85 ng/ml, with a total assay time of 2 hours 30 minutes. Kit components are stored at +4 °C or -20 °C as instructed and should not be used past the expiration date. Reported recovery values are 88-102 % for serum, 90-98 % for EDTA plasma and 90-102 % for heparin plasma. Dilution linearity ranges across 1:2 to 1:16 dilutions, with serum values of 82-96 %, 83-98 %, 81-99 % and 93-101 %, EDTA plasma values of 88-101 %, 86-95 %, 90-102 % and 80-93 %, and heparin plasma values of 80-91 %, 82-90 %, 95-104 % and 79-95 %.
